Antifungal activity and biocompatibility on fibroblast cells of Sea Urchin (Diadema setosum) gonad lipid extract based PVA (polyvinyl alcohol) hydrogel

In this study, a hydrochar hydrogel was prepared using a polyvinyl alcohol/polyvinyl pyrrolidone (PVA/PVP) polymer network combined with EO. The essential oil emulsion was bonded to the hydrochar-enriched PVA/PVA hydrogel (H*PPg) using a lyophilizer. Some chemical properties and antifungal activity of the hydrochar-bound PVA/PVP gel combined with EO (H*PPg@EO) against Candida yeasts were investigated. The thermal properties, characteristics, and topographic images of the H*PPg and H*PPg@EO gels were evaluated using differential scanning calorimetry-thermogravimetric analysis (DSC-TGA), Fourier transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM), respectively. The antifungal properties of H*PPg and H*PPg@EO hydrogels were evaluated against Candida albicans and Candida parapsilosis. GC-MS analysis on Citrus sinensis revealed that the dominant component was limonene with a ratio of 73.34%, while FTIR analysis confirmed the successful integration of the essential oil into the pVA/PVP matrix containing hydrochar. The more negative zeta potential of -10.8±1.1 mV and higher conductivity value of 0.738 mS/cm of H*PPg@EO indicated that it formed a more stable system compared to H*PPg. SEM observations showed that the porous network structure was preserved and the surface heterogeneity increased after EO loading. Biological evaluations revealed that H*PPg@EO exhibited significant inhibition zones and high inhibition percentages against C. albicans (15.6 ± 2.1 mm) and C. parapsilosis (14.2 ± 2.1), while the H*PPg form did not show antifungal activity. These findings highlight the important role of monoterpenes, including limonene, in antifungal activity and demonstrate the advantage of sustained release within the hydrogel network. Consequently, H*PPg@EO is confirmed as a promising and innovative biomaterial with potential applications in controlled-release systems, functional foods, biomedical fields, and antimicrobial coatings.
